Wearable BIOsensor based on ISOthermal nucleic acid amplification for PAThogen detection from skin wounds

 

 

Objectives

Briefly, the scope and objective of this project is to design, manufacture and demonstrate the applicability of a wearable point-of-care system based on a flexible substrate with a microfluidic system for the isothermal amplification of gene fragments corresponding to the most representative pathogens present in skin lesions.

The project objectives of Bio-Iso-Pat project are:

O1. Design, fabrication and optimization of the point-of-care system, which will be comprised of i) the flexible substrate, chemically modified with single stranded DNA primers (ssDNA) which will capture the gene fragments corresponding to the pathogens (bacterial, fungal) and ii) the microfluidic system which will act as a reaction chamber for the isothermal amplification.

O2. Design, testing and optimization of molecular biology protocols, which will explore DNA extraction-free solutions for nucleic acid amplification. Moreover, this objective aims to ensure the correct primer design and an appropriate fluorescent labeling protocol, in order to avoid false-positive and false-negative results.
